Specimen Collection
4 mL Adult Heparin Blood

Dark green top Heparin tubes are required ,

light green top PST Heparin tubes(containing gel) are NOT suitable.

1 mL Paediatric Heparin Blood

Dark green top Heparin tubes are required ,

light green top PST Heparin tubes(containing gel) are NOT suitable.

• Tested Mon - Fri only. Blood samples must arrive at the laboratory before midday Friday or on the day prior to public holiday.

 

For samples sent to LabPlus from outside the Auckland region:

• Must be pre-arranged with cell marker staff. Sample collection as above , transport at room temperature.
• Sample age limit for testing to be completed is 24 hours.

Diagnostic Use and Interpretation

1. To determine the phenotypic severity of DiGeorge Syndrome.
2. Selected bone marrow transplant recipients to determine engraftment.
3. Novel Immune deficiency disorders.
4. Selected rheumatoid arthritis patients.
5. Selected HIV patients.

CD45RA and CD62L are used to identify naive T-cell subsets, CD45RO and CD62L are used to identify memory T-cell subsets. The following phenotypes have been described in literature:

CD4+ T helper naive cells:                CD4+CD45RA+CD62L+

CD4+ T helper memory cells:           CD4+CD45RO+CD62L+
                                                                CD4+CD45RO+CD62L-
                                                                CD4+CD45RA+CD62L- (very small subset)

CD8+ T cytotoxic naive cells:            CD8+CD45RA+CD62L+

CD8+ T cytotoxic memory cells:        CD8+CD45RO+CD62L+
                                                                 CD8+CD45RO+CD62L-

                                                                 CD8+CD45RA+CD62L-
