Specimen Collection

The test is only performed Monday - Friday 0800:1700 hrs.

Separate sample for Soluble CD25; cannot be shared with Chemistry.

Internal : Immediately spin and separate plasma to 2 x 1ml aliquots. Hand deliver spun and separated Heparin plasma to Coagulation Lab. Freeze at -20c. 

External Laboratories must FREEZE the plasma within 24-hours of collection and send 2 x 1ml Heparin Plasma aliquots to LabPlus frozen.

Requests for Soluble CD25 must be arranged through LabPlus Haematologist who will advise when results will be available.


Assay Method

Samples were tested using Quantikine R Human   CD25/IL-2Ra Immunoassay which is a 3.5 hour solid phase ELISA designed to measure soluble human IL-2Ra in cell cultures ,serum and Heparin plasma. It contains NS0 expressed recombinant human IL-2Ra and antibodies raised against the recombinant factor. Results obtained using natural Human IL-2Ra showed linear curves that were parallel to the standard curves obtained using the Quantikine R   kit standard indicating this kit can be used to determine relative mass values for natural human IL-2Ra. This assay employs the quantitative sandwich enzyme immunoassay technique .A monoclonal antibody specific for human IL-2Ra has been pre coated onto a microplate. Standards, samples, and conjugate are pipetted into the wells and any IL-2Ra present is sandwiched by the immobilized antibody and the enzyme linked polyclonal antibody specific for human IL-2Ra.


Diagnostic Use and Interpretation

Elevated serum level of IL2 Receptor alpha ( IL-2Ra ) is important in the diagnosis of HLH (Haemophagocytic Lymphohistiocytosis), which can be used to monitor treatment and to evaluate disease activity.

Specimen Transport Instructions for Referring Laboratories
